Kinetics of Electron Returns in Successive Two-Photon DNA Repair by (6-4) Photolyase

چکیده
(6-4) photolyase is a flavoenzyme that catalyzes the chemically challenging repair of carcinogenic photoproduct lesions in DNA using sunlight. Nature may have overcome challenge by combining two distinct successive photoreactions, each starting with electron transfer from an excited flavin to lesion. We herewith report transient absorption study returns following chemical transformations successfully dissected kinetics finalize first and second photoreactions (∼40 μs ∼200 ns, respectively) Xenopus laevis, corroborating detailing two-photon reaction model.

(6-4) photolyase: light-dependent repair of DNA damage.
DNA photolyase represents a phenomenal class of DNA repair enzymes in that it harvests the light energy to repair DNA lesions caused by ultraviolet light. Mother Nature evolves two types of photolyases, one specific for repairing cyclobutane pyrimidine dimers and the other for pyrimidine-(6-4)-pyrimidone photoproducts. Together, these two kinds of DNA photolesions account for the majority of ul...

Background: Exposure to ionizing radiation is known to induce oxidative stress followed by damage to critical biomolecules like lipids, proteins and DNA through radiolysis of cellular water. Since radiation has been widely used as an important tool in therapy of cancer, the detailed investigation regarding the DNA damage and repair kinetics would help to predict the radiation sensitivity of cel...

Cyclobutane pyrimidine dimers are the major DNA photoproducts produced upon exposure to UV radiation. If left unrepaired, these lesions can lead to replication errors, mutation, and cell death. Photolyase is a light-activated flavoenzyme that binds to pyrimidine dimers in DNA and repairs them in a reaction triggered by electron transfer from the photoexcited flavin cofactor to the dimer. Using ...

The reaction mechanism of Xenopus (6-4) photolyase was investigated using several mutant enzymes. In the active site, which is homologous between the cis,syn-cyclobutane pyrimidine dimer and (6-4) photolyases, four amino acid residues that are specific to (6-4) photolyase, Gln(288), His(354), Leu(355), and His(358), and two conserved tryptophans, Trp(291) and Trp(398), were substituted with ala...
